Dave Hennigan, the former Rochdale Lib Dem Agent, has put a formal complaint into Parliamentary authorities about Simon Danczuk's staff expenses.
Mr Hennigan has called for an investigation into the employment of Karen Danczuk (employed as Ms Burke).
Mr Hennigan says he expects the Independent Parliamentary Standards Authority (IPSA) to launch an investigation that "could lead to formal action against Mr Danczuk".
The Lib Dems are also asking asking for evidence that the recruitment process of Mr Danczuk's new parliamentary aide, Nasreen Nazir, was transparent to avoid allegations of cronyism.
Mr Hennigan said: "These latest revelations expose the hypocrisy of Danczuk and his team."
The complaint has been backed by Liberal Democrat Parliamentary Spokesperson for Rochdale, Councillor Andy Kelly.
Councillor Kelly said: "There are so many questions for Mr Danczuk to answer. It is only right that we go by the proper and due process.
"Staffing expenses are not some sort of slush fund of MPs to employ their mates and spouses. They are there to ensure a Member of Parliament provides the service that local people need and deserve. After all, it's our tax that is paying for them.
"I look forward to a thorough and robust investigation."
Mr Danczuk did not respond to a request for comment.
